INDICATION

SGIndrop D 5000 IU Caps is a vitamin D3 supplement indicated for the prevention and treatment of vitamin D deficiency and insufficiency, including in patients with osteoporosis, osteomalacia, secondary hyperparathyroidism, limited sun exposure, malabsorption, and as maintenance therapy following high-dose loading regimens.

MECHANISM OF ACTION

Cholecalciferol (vitamin D3) is the natural form of vitamin D produced in the skin under ultraviolet B light. After oral ingestion, it is hydroxylated in the liver to 25-hydroxyvitamin D3 (calcidiol) and subsequently in the kidney to 1,25-dihydroxyvitamin D3 (calcitriol), the biologically active hormone. Calcitriol acts on vitamin D receptors to enhance intestinal calcium and phosphorus absorption, regulate parathyroid hormone secretion, support bone mineralization, and modulate immune function. The 5000 IU daily dose is used for moderate deficiency correction and long-term maintenance in deficient patients.

DOSAGE AND ADMINISTRATION

      Vitamin D deficiency: 5000 IU once daily for 8 to 12 weeks, then reassess serum 25-OH vitamin D

      Maintenance in deficient patients: 5000 IU daily or as prescribed based on blood levels

      Take with a fatty meal to improve absorption

      Always follow the dosage and monitoring schedule prescribed by your doctor

SIDE EFFECTS

Common:

      Generally, well tolerated at 5000 IU daily

      Mild nausea or stomach discomfort

      Headache

      Constipation

Less common:

      Hypercalcemia with excessive or prolonged use: thirst, frequent urination, weakness

      Kidney stones with prolonged hypercalcemia

      Hypercalciuria

      Fatigue or confusion at toxic doses

NOT RECOMMENDED IN

      Hypercalcemia or hypercalciuria

      Known hypersensitivity to cholecalciferol

      Severe renal impairment without specialist guidance

DRUG INTERACTIONS

Thiazide diuretics combined with vitamin D increase hypercalcemia risk. Orlistat, cholestyramine, and mineral oil reduce vitamin D absorption; take separately. Antiepileptics including phenytoin, carbamazepine, and phenobarbitone increase vitamin D catabolism, requiring higher supplementation. Digoxin toxicity risk increases with hypercalcemia from excess vitamin D.
