INDICATION
Pelton-V is indicated for the treatment and prevention of nausea and vomiting of various origins, including post-operative nausea, chemotherapy-induced nausea, and nausea associated with gastrointestinal disorders. It is also used to manage gastroparesis, reflux oesophagitis, non-ulcer dyspepsia, and bloating caused by delayed gastric emptying.
MECHANISM OF ACTION
Pelton-V contains Domperidone Maleate, a peripheral dopamine D2 receptor antagonist. It blocks dopamine receptors in the chemoreceptor trigger zone (CTZ) of the brain, thereby suppressing the nausea and vomiting signal. In the gastrointestinal tract, it enhances the coordinated contractions of the stomach and upper intestine, accelerating gastric emptying and reducing the sensation of bloating and fullness.
DOSAGE AND ADMINISTRATION
ā¢Ā Ā Ā Ā Ā Adults and adolescents (ā„12 years, ā„35 kg): 10 mg up to three times daily, preferably 15ā30 minutes before meals.
ā¢Ā Ā Ā Ā Ā Maximum dose: 30 mg per day.
ā¢Ā Ā Ā Ā Ā Treatment should not exceed 12 weeks without medical review.
ā¢Ā Ā Ā Ā Ā Take with a glass of water; do not crush or chew.
SIDE EFFECTS
ā¢Ā Ā Ā Ā Ā Dry mouth, headache, or abdominal cramps.
ā¢Ā Ā Ā Ā Ā Raised prolactin levels which may cause breast tenderness, galactorrhoea, or menstrual irregularities.

NOT RECOMMENDED IN
Contraindicated in patients with gastrointestinal haemorrhage, mechanical obstruction, or perforation. Not recommended in patients with moderate to severe hepatic impairment, known cardiac arrhythmias, or a prolactin-secreting pituitary tumour. Avoid in patients over 60 years of age without careful assessment.
DRUG INTERACTIONS
Domperidone interacts with QT-prolonging drugs (e.g., antifungals like ketoconazole, certain antibiotics, and antiarrhythmics) so concurrent use is contraindicated. It may also interact with anticholinergic drugs, opioid analgesics, and antacids.
