INDICATION

Oline Plus 3mg+25mg Caps is a fixed-dose combination of an atypical antipsychotic and a selective serotonin reuptake inhibitor (SSRI), indicated for the treatment of depressive episodes associated with bipolar I disorder and treatment-resistant depression. It is prescribed when adequate trials of standard antidepressant therapy have been insufficient.

MECHANISM OF ACTION

Olanzapine is an atypical antipsychotic that antagonizes dopamine D2 and serotonin 5-HT2A receptors, stabilizing mood, reducing agitation, and preventing manic switching in bipolar depression. Fluoxetine is a selective serotonin reuptake inhibitor (SSRI) that blocks the serotonin transporter (SERT), increasing synaptic serotonin availability and producing antidepressant effects. The combination provides synergistic antidepressant activity: olanzapine modulates dopaminergic and serotonergic tone while fluoxetine directly augments serotonergic neurotransmission, making the combination effective in treatment-resistant and bipolar depression where monotherapy is insufficient.

DOSAGE AND ADMINISTRATION

•      Adults: 1 capsule once daily in the evening
