INDICATION
Lovir Eye Ointment is an antiviral ophthalmic preparation indicated for the treatment of herpes simplex virus keratitis (dendritic ulcer), a potentially sight-threatening corneal infection caused by HSV-1 or HSV-2.
MECHANISM OF ACTION
Acyclovir is a nucleoside analogue that requires viral thymidine kinase for initial phosphorylation to acyclovir monophosphate. Cellular kinases then convert it to the active triphosphate form, which competitively inhibits viral DNA polymerase and acts as a chain terminator when incorporated into viral DNA. Because acyclovir requires viral thymidine kinase for activation, it selectively targets virus-infected cells while sparing healthy tissue.
DOSAGE AND ADMINISTRATION
● Apply a 1cm ribbon of ointment into the lower conjunctival sac 5 times daily at approximately 4-hour intervals
● Continue treatment for at least 3 days after complete healing of the ulcer
● Typical treatment duration is 7 to 14 days
● Vision may be temporarily blurred after application
● Do not wear contact lenses during treatment
SIDE EFFECTS
Common:
● Temporary blurring of vision after application
● Mild stinging or burning on instillation
Less common:
● Superficial punctate keratopathy in rare cases
● Local allergic reaction including eyelid edema
NOT RECOMMENDED IN
● Known hypersensitivity to acyclovir or valacyclovir
● Do not use for bacterial, fungal, or non-herpetic viral eye infections
DRUG INTERACTIONS
No clinically significant drug interactions with ophthalmic acyclovir. If using other eye preparations concurrently, apply the ointment last.
