INDICATION

Lodopin-V 5/80mg is a fixed-dose antihypertensive combination tablet indicated for the management of essential hypertension in adult patients whose blood pressure is not adequately controlled on monotherapy. It combines amlodipine, a calcium channel blocker, with valsartan, an angiotensin II receptor blocker (ARB), to provide complementary and additive blood pressure-lowering effects. This combination is effective in reducing the risk of cardiovascular events such as stroke and myocardial infarction associated with uncontrolled hypertension.

MECHANISM OF ACTION

Amlodipine blocks voltage-dependent L-type calcium channels in vascular smooth muscle and cardiac cells, causing vasodilation and a reduction in peripheral vascular resistance. Valsartan selectively blocks AT1 angiotensin II receptors, preventing angiotensin II from exerting its vasoconstrictive and aldosterone-stimulating effects. Together, these mechanisms result in significant and sustained blood pressure reduction.

DOSAGE AND ADMINISTRATION

•      One tablet of 5/80mg once daily at the same time each day.

•      May be taken with or without food.

•      Blood pressure response should be evaluated after 2–4 weeks of therapy.

•      Higher strength combinations are available if the target BP is not achieved.

•      Do not abruptly discontinue antihypertensive therapy.

NOT RECOMMENDED IN

Lodopin-V 5/80mg is contraindicated in patients with known hypersensitivity to amlodipine, valsartan, or any excipient. It is not recommended during the second and third trimesters of pregnancy due to the risk of fetal harm. Patients with severe hepatic impairment, biliary cirrhosis, or cholestasis should avoid valsartan.

DRUG INTERACTIONS

Co-administration of Lodopin-V with other antihypertensives or diuretics may produce additive blood pressure lowering. NSAIDs can reduce the antihypertensive effect of valsartan and may worsen renal function. Potassium-sparing diuretics or potassium supplements combined with valsartan increase the risk of hyperkalemia.
