INDICATION

Lodopin-V 5/160mg Tab is a fixed-dose combination antihypertensive containing a calcium channel blocker and an angiotensin receptor blocker (ARB). It is indicated for the treatment of hypertension in patients not adequately controlled on monotherapy, reducing the need to take two separate tablets.

MECHANISM OF ACTION

Amlodipine blocks L-type voltage-gated calcium channels in vascular smooth muscle and cardiac tissue, reducing calcium influx and causing peripheral vasodilation, which lowers blood pressure and reduces cardiac afterload. Valsartan selectively blocks the angiotensin II type 1 (AT1) receptor, preventing angiotensin II-mediated vasoconstriction, aldosterone release, and sodium retention. Together, these complementary mechanisms produce additive blood pressure reduction with a favorable tolerability profile.

DOSAGE AND ADMINISTRATION

•      Adults: 1 tablet once daily

•      May be taken with or without food

•      Dose adjustment may be required in severe renal or hepatic impairment; consult your doctor

•      Always follow the dosage and blood pressure monitoring schedule prescribed by your physician

SIDE EFFECTS

Common:

•      Peripheral oedema (ankle swelling) from amlodipine

•      Headache and dizziness

•      Fatigue

•      Flushing

Less common:

•      Hyperkalemia (elevated potassium) from valsartan component

•      Elevated creatinine or renal impairment

DRUG INTERACTIONS

Potassium supplements, potassium-sparing diuretics, and other ARBs increase hyperkalemia risk with valsartan. NSAIDs reduce the antihypertensive effect and may worsen renal function. CYP3A4 inhibitors such as ketoconazole and clarithromycin increase amlodipine plasma levels. Simvastatin doses should be limited to 20mg/day when combined with amlodipine due to increased myopathy risk.
