INDICATION
Gelofusine 500ML is a colloid plasma volume expander indicated for the treatment of hypovolemia due to acute blood loss, burns, trauma, or surgery. It is used when crystalloid solutions alone are insufficient to restore circulating blood volume and maintain hemodynamic stability.
MECHANISM OF ACTION
Succinylated gelatin 4% is a modified gelatin-based colloid that generates oncotic pressure within the intravascular compartment. When infused intravenously, it draws fluid from the interstitial space into the blood vessels, expanding plasma volume rapidly and supporting blood pressure and cardiac output. Each 500ml of Gelofusine provides a volume expansion roughly equivalent to the volume infused, with effects lasting 3 to 4 hours. It also contains sodium chloride for electrolyte balance.
DOSAGE AND ADMINISTRATION
● Administered intravenously under medical supervision only
● Initial dose: 500ml to 1000ml infused at a rate determined by clinical urgency
● In acute hemorrhagic shock, rapid infusion rates may be required
● Monitor vital signs, urine output, and hemodynamic parameters throughout
● Maximum daily dose: 20ml/kg body weight unless clinically justified
● Warm to body temperature before infusion if administered in large volumes
SIDE EFFECTS
Common:
● Mild allergic reactions including urticaria or flushing
● Transient tachycardia during rapid infusion
Less common:
● Anaphylactoid reactions in rare cases
● Dilutional coagulopathy with large-volume resuscitation
● Fluid overload in patients with cardiac or renal impairment
NOT RECOMMENDED IN
● Known hypersensitivity to gelatin-based colloids
● Severe cardiac failure with pulmonary edema
● Hypervolemia
● Severe coagulation disorders
