INDICATION
Admore Drops 4000 IU is a vitamin D3 supplement in oral drop form indicated for the prevention and treatment of vitamin D deficiency in adults and children, including those with limited sun exposure, malabsorption syndromes, and conditions such as rickets, osteomalacia, and secondary hyperparathyroidism. It is particularly suitable for infants and young children, the elderly, and patients who are unable to swallow solid dosage forms. MECHANISM OF ACTION
Cholecalciferol (vitamin D3) is the natural form of vitamin D synthesized in the skin upon ultraviolet B light exposure. After oral ingestion, it is hydroxylated in the liver to 25-hydroxyvitamin D3 (calcidiol), and subsequently in the kidney to the biologically active 1,25-dihydroxyvitamin D3 (calcitriol). Calcitriol acts on vitamin D receptors in the intestinal epithelium to enhance calcium and phosphorus absorption, in the kidney to reduce urinary calcium excretion, and in bone to support mineralization. It also regulates parathyroid hormone secretion and modulates immune function and cell differentiation.
DOSAGE AND ADMINISTRATION
• Infants and children: 400 to 1000 IU daily (1 to 2.5 drops) for prevention; therapeutic doses as prescribed by pediatrician
• Adults: 1000 to 4000 IU daily (2.5 to 10 drops) based on serum 25-OH vitamin D levels
• Administer directly into the mouth or mixed with a small amount of food or drink
• Always follow the dosage prescribed by your doctor based on blood vitamin D levels and clinical need
SIDE EFFECTS
Common:
• Generally, well tolerated at recommended doses
• Mild nausea or stomach discomfort at higher doses
• Constipation
• Headache
Less common:
• Hypercalcemia with excessive prolonged use: thirst, polyuria, weakness, confusion, and cardiac effects
• Hypercalciuria and kidney stone formation with very high doses
• Metastatic calcification with toxic doses

DRUG INTERACTIONS
Thiazide diuretics combined with high-dose vitamin D increase hypercalcemia risk; monitor serum calcium. Antiepileptics including phenytoin, carbamazepine, and phenobarbitone increase vitamin D catabolism. Digoxin toxicity risk increases with hypercalcemia from excess vitamin D.
