INDICATION
Acylex Cream is a topical antiviral preparation indicated for the treatment of recurrent herpes labialis (cold sores) caused by herpes simplex virus type 1 (HSV-1) on the lips and face in adults and children 12 years of age and older. It reduces the duration and severity of cold sore episodes when applied at the first sign of recurrence, such as tingling, itching, or burning before a blister appears. It is also used for the management of superficial herpes simplex skin infections in immunocompetent patients.
MECHANISM OF ACTION
Acyclovir is a synthetic purine nucleoside analogue that is selectively phosphorylated in herpes-infected cells by viral thymidine kinase to its monophosphate form, and then further phosphorylated by host cellular kinases to acyclovir triphosphate. This active metabolite competitively inhibits herpes viral DNA polymerase and is incorporated into viral DNA, causing chain termination. The high selectivity for viral thymidine kinase results in a 40 to 100 times higher intracellular concentration of acyclovir triphosphate in infected cells compared to uninfected cells, producing antiviral activity with minimal toxicity to host cells.
DOSAGE AND ADMINISTRATION
• Apply a thin layer of cream to the affected area 5 times daily, approximately every 4 hours, for 5 days
• Begin treatment at the first sign of recurrence (tingling, itching, or burning) for maximum benefit
• Wash hands before and after application; avoid getting cream in or near the eyes
• Do not apply inside the mouth or on mucous membranes; always follow the advice of your doctor or pharmacist
SIDE EFFECTS
Common:
• Mild burning or stinging on application
• Itching at the application site
• Skin dryness or flaking at the application site
• Mild erythema (redness)
Less common:
• Contact dermatitis or allergic reaction in sensitive individuals
• Angioedema (rare)
• Oedema at the application site
• Desquamation with prolonged or excessive use
NOT RECOMMENDED IN
• Known hypersensitivity to acyclovir, valacyclovir, or propylene glycol (excipient)
